  • Monitoring of Apoprotein-Lipid-Assemblies to Nanodiscs

A Nanodisc is a soluble nanoscale non-covalent assembly of phospholipid bilayers and a genetically engineered  "membrane scaffold protein" (based upon the sequence of human serum apolipoprotein A-I). The phospholipid associates as a bilayer fragment while to molecules of apo-protein wrap around the edges of the discoidal structure in a belt-like configuration, covering the hydrophobic alkyl moiety of the lipid-molecules. The  feature of nanodiscs is that they self-assemble integral membrane proteins make them water soluble at a single molecule level, which makes this approach superior over liposomes or detergent micelles in terms of size and stability. Here we present the radius distribution signatures of a successful integration of a protein after self-assembly as an indicator for optimized reaction parameters. As a control we show the signature of a a detergent inhibited self-assembly.
